血浆中铂的微量进样-电感耦合等离子体质谱的快速测定法

【摘要】 目的建立微量进样-电感耦合等离子体质谱(ICP-MS)测定血浆中铂元素浓度的快速检测方法。方法血浆样品不经过稀释利用高效液相色谱仪自动进样器直接进样20μl,进行ICP-MS,经外标法测定血浆中铂元素浓度。结果血浆中铂元素浓度在0.5~500.0 ng/ml的线性范围内,所得回归方程为y=0.0 447 322x-3.995 22,r=0.999 56。该方法的检出限为0.16 ng/ml,定量下限为0.52 ng/ml,RSD为0.82~5.41%,回收率为100.7%~106.7%。结论该方法灵敏度高,方法检出限低,重复性好,取样量少,操作简单快速,适用于血浆中铂元素浓度临床大批量样品的分析。

【Abstract】 Objective To establish a method for detecting platinum in plasma by micro-volume injection and inductively coupled plasma-mass spectrometry(ICP-MS). Methods The plasma samples without dilution were directly added to ICP-MS by using an automatic sampler(20 μl) of high performance liquid chromatography(HPLC). Results Within the linear range of0.5-500 ng/ml,the regression equation of platinum was y=0.0 447 322 x-3.995 22,The correlation coefficient was 0.999 6. The detection limit was 0.16 ng/ml. The determination limit was 0.52 ng/ml. The within-run relative standard deviations(RSDs) of this methods were 0.82%-5.41%,and the average recoveries were 100.7%-106.7%. Conclusion With the advantages of high sensitivity,good repeatability,little sample volume and easy operation,this method is applicable to the determination of platinum in plasma.
