酒精有害使用对昼夜节律相关基因mRNA表达及睡眠行为的影响

【摘要】 目的:探讨酒精有害使用(HUA)对昼夜节律相关基因mRNA表达及其对睡眠行为的影响。方法:收集网络招募的22例HUA者(HUA组)及21名健康对照者(对照组)的基本信息,并对入组者进行匹兹堡睡眠质量指数(PSQI)评估、多导睡眠监测(PSG)及外周血白细胞中昼夜节律相关基因[钟基因(Clock)、周期基因1(Per1)、Per2、Per3、大脑和肌肉ARNT样蛋白-1基因(Bmal1)] mRNA表达测定;对结果进行组间比较,分析昼夜节律相关基因mRNA表达及其对睡眠行为的影响。结果:HUA组Clock、Per3、Bmal1基因mRNA表达明显低于对照组(P<0.05或P<0.01);PSQI总分及睡眠效率评分明显高于对照组(P均<0.05);PSG观察指标中,总睡眠时间、睡眠效率、非快速眼动2期睡眠及快速眼动期睡眠时间明显低于对照组(P均<0.05),睡眠潜伏期和入睡后清醒时间明显高于对照组(P均<0.05)。HUA组Clock mRNA表达与饮酒年限呈负相关(r=-0.434,P<0.05)。结论:HUA者Clock、Per3、Bmal1基因表达下降,并且出现睡眠时间、效率、睡眠连续性的下降和睡眠起始困难。

【Abstract】 Objective:To investigate the effects of harmful use of alcohol(HUA) on the mRNA expression of circadian rhythm related genes and sleep behavior. Method:Basic information of recruited online HUA subjects(HUA group, n=22) and healthy controls(control group, n=21) were collected, the Pittsburgh Sleep Quality Index(PSQI) and polysomnography(PSG) were assessed and related circadian genes in peripheral blood leukocytes [Clock genes(Clock),cycle gene 1(Per1),Per2,Per3,brain and muscles to ARNT sample protein 1 gene(Bmal1)] mRNA expression were determinated.The results were compared between the two groups, the mRNA expression of circadian rhythm related genes and their effects on sleep behavior were analyzed. Results:The mRNA expressions of Clock, Per3 and Bmal1 in the HUA group were significantly lower than those in the control group(P<0.05 or P<0.01).The total score and sleep efficiency score of PSQI were significantly higher than those of control group(all P<0.05).The total sleep time, sleep efficiency, non-rapid eye movement(REM) phase 2 sleep and REM sleep time were significantly lower than those in the control group(all P<0.05),and the sleep latency and waking time after sleep were significantly higher than those in the control group(all P<0.05).There was a negative correlation between Clock mRNA expression and drinking years in the HUA group(r=-0.434,P<0.05). Conclusion: The expression of Clock, Per3 and Bmal1 genes in the HUA people decreased, and there are also decreases in sleep time, efficiency and sleep continuity, and difficulty in sleep initiation.
