Study on Protective Agent to Improve the Stability of Leather Protease
【摘要】 酶制剂在皮革行业的应用越来越广泛,为提高皮革用蛋白酶的应用性能,促进清洁化酶制剂在皮革工业的推广,本文研究了醇类、糖类和有机溶剂作为保护剂对皮革用蛋白酶热稳定性的影响。结果表明,添加山梨醇、葡萄糖和甘油可显著提高蛋白酶的热稳定性,并通过响应面试验优化得到复合保护剂配方为13%山梨醇和25%葡萄糖,综合考虑应用于皮革脱毛后的操作与成本问题,最终选择添加25%葡萄糖作为皮革用蛋白酶的保护剂。添加25%葡萄糖的蛋白酶在50℃保存6 h,酶活保存率为85.73±1.28%,较对照提高了61.83%。且蛋白酶的半衰期显著提高,当温度大于40℃时添加保护剂与不添加保护剂的蛋白酶相比半衰期分别提高了1.00倍、2.60倍、1.25倍和2.13倍。
【Abstract】 Enzyme preparations are widely applied in the leather industry. In order to improve the application performance of leather proteases and promote the popularization of cleaning enzyme preparations in leather industry, the effects of alcohols, sugars and organic solvent protective agents on the thermal stability of leather proteases were studied. The results showed that the addition of sorbitol,glucose and glycerol could significantly improve the thermal stability of the protease, and the compound protective agent formula was optimized by response surface test to be 13% sorbitol and 25%glucose. The protective agent for the leather protease was 25% glucose, considered the operation and cost of the enzyme in leather depilation. After the protease with 25% glucose was stored at 50℃ for 6 h, the retention rate of enzyme activity was 85.73±1.28%, which was 61.83% higher than that of the control. The half-life of the protease increased significantly, when the temperature was higher than40℃. The half-life of the protease with the protection agent was 1.00 times, 2.60 times, 1.25 times and 2.13 times higher than that of the protease without the protection agent.
